新一代的荧光染色展开了荧光技术的新前程
荧光技术及其应用 -- 经典荧光染色 :
荧光具有两种特殊的光谱 : 1) 激发光谱(波长和光吸收量) ; 2) 发射光谱(波长和光发射量)
这两种能量的吸收和发射是荧光技术独特的分子特征. 因此, 荧光测定被选择用于极其敏感, 高度特异性,简便易行, 成本低等有利特征来与其他测定技术相比较. 实际上, 荧光测定比普通的测定方法要敏感 10-1000倍. 此技术广泛地被医学生物学领域所接受, 并已成为极其有价值的定性及定量分析测定的有力工具.
荧光测定特点为 :
1) 可用于很小的样本测定 ;
2) 不易受测定样品及环境等多方面因素干扰 ;
3) 不必稀释样品浓度, 只需增强荧光光度即可做各种不同浓度的测定 ;
4) 大部分荧光测定被用于自动大量筛选样品的检测.

HiLyte FluorTM荧光染色剂是一系列高质量荧光标记的染色剂并可扩至所有可见光谱. 大部分HiLyte FluorTM荧光染色剂具有良好的水溶性, 因此蛋白的联结可应用各种非有机溶剂来进行, 并且此种蛋白联结在贮存时很少产生沉淀.
这些新一代的染色剂与经典荧光染色剂相比较, 具有较强的标记性能.
此类荧光染色剂及其联结物的优势为 :
· 可用于各种反应形式，通用范围广
· 标记物具有强烈的荧光光度，与经典荧光剂FITC, TAMRA及ROX相比
· 具有更佳的照相稳定性，与经典荧光剂FITC, Cy3及Cy5相比
· 具有与基本激发波长相匹配的吸收光谱， 如488 nm, 555 nm, 633 nm 及 647 nm
· 染色剂及其标记物可适用于几种截然不同的荧光颜色
· 在不同的pH范围同样具有强烈的荧光
· 容易与其它产品联结

HiLyte FluorTM 488 用于标记糖蛋白，如HRP, 与绿色荧光FITC比较 :
- 标记物荧光亮度极其明显提高 ;
